为了探讨cyclinE，cdk2在乳腺癌细胞周期进程中的作用，利用DNA 重组技术构建了cyclinE以及cdk2的反义RNA真核表达载体。通过磷酸钙 共沉淀法，将重组质粒转入人乳腺癌细胞系Bcap 37（简称B37，经G418 筛选得到各质粒转染的阳性克隆分别命名为NLE，NLK2. 利用neo基因特异的引物，通过PCR检测确定在NLE，NLK2中均含 有重组质粒，而且Western blot结果也表明在NLE中的cyclinE，在NLK2 中的CDK2表达水平明显下降。 建立起细胞模型后，为了探讨cy
