线粒体是大多数真核细胞进行能量转换的重要细胞器,已有许多研究证明线粒体和衰老有关。近年有报道通过离心的方法可将有活性的线粒体转入受体细胞,但对于转移的线粒体缺乏严谨的质量控制方案。本研究基于这一现状,设计了严格而简便的线粒体质量控制方法,并且对实验步骤进行了改良。本文利用蔗糖梯度密度离心法提取线粒体,RIPA裂解提取线粒体蛋白质和DNA,分别通过蛋白免疫印迹(WB)和聚合酶链式反应(PCR)检测线粒体纯度;而后使用JC-1染液对提取的线粒体样品进行染色,荧光显微镜观察结果表明该方法提取的线粒体是完整而且有活性的;最后使用活线粒体荧光探针Mito-Tracker Green孵育后的细胞作为线粒体供体,通过离心转移至受体细胞,贴壁后用活线粒体荧光探针Mito-Tracker Red孵育,荧光显微镜观察红绿荧光信号重合表明活线粒体成功转移入受体细胞。

Mitochondria are important organelles for the production of energy in most eukaryotic cells, and many studies have shown that mitochondria are associated with aging. In recent years, it has been reported that active mitochondria can be transferred into cells by simple centrifugation, but the quality control on mitochondria is not strict enough. This study focused on the initial improvement of mitochondrial quality control steps. Mitochondria were extracted by sucrose density gradient centrifugation, total protein and DNA samples were extracted and detected by Western blotting and PCR, respectively, which proved that high-purity mitochondria are extracted; mitochondrial samples were stained with JC-1, fluorescence results indicated that the mitochondria extracted by the method were intact and active; the cells incubated with the live mitochondrial fluorescent probe Mito-Tracker Green were used to extract mitochondria, transferred to the same recipient cells by simple centrifugation. After incubating the recipient cells with the live mitochondrial fluorescent probe MitoRed, fluorescence microscopy observation of red-green fluorescence signal coincidence indicates that the live mitochondria were successfully transferred into the cells.
