研究目的:SO2被认为是大气污染物的主要成分之一,对环境和人类的健康危害很大。SO2被吸入体内之后,在体液中转化为它的衍生物—亚硫酸钠(Na2SO3)和亚硫酸氢钠(NaHSO3),通过血液循环分布全身,从而对其他组织、器官产生影响 。实验室前期研究显示,SO2和运动双重刺激不仅会抵消运动降压和改善心功能的健康效应,而且会使血压升高,心脏的收缩功能下降,对心脏功能产生更为不利的影响。心的功能不仅取决于心肌本身的收缩功能,还与心肌胶原纤维有密切的联系。心肌胶原纤维表达异常,以及心肌纤维化等临床表现与心功能下降有关。由此推测,SO2污染环境引起大鼠心肌收缩功能改变可能与心肌形态学和心肌胶原蛋白含量的变化有关。本实验拟对SO2吸入和适宜运动双重作用下心肌胶原纤维及其局部RAS系统的核心成员AngⅡ受体(Angiotensin II Type 1 Receptor,AT1R)含量以及结缔组织生长因子(Connective tissue growth factor,CTGF)的表达水平的变化进行研究,探讨SO2吸入和适宜运动对心肌胶原纤维的影响及可能机制。研究方法:24只SD大鼠随机分为空白对照组(RG)、单纯运动组(EG)、单纯SO2污染组(SRG)、SO2污染运动组(SEG),每组6只。SRG和SEG置于SO2污染环境中(10mg/m3),采用动态吸入染毒法对其进行SO2染毒。运动组大鼠进行跑轮运动(8m/min,1h/d,共4周)。每周测试大鼠体重二次,比较4组大鼠体重变化情况。最后一次染毒后大鼠禁食24h,称重(g),用2%戊巴比妥纳(3mL/kg)麻醉;剖腹,开胸迅速取出心脏,剪除大血管,冰盐水冲洗,滤纸吸干水分,称全心重量(HW)后,立即在冰生理盐水中迅速沿房室交界处去除左右心房以及大血管,去除右心室游离壁,称取余下的左心室重量(LVM),分别计算心脏重量指数(HBI)[HBI=心脏重量(g)/体重(g) ×1000]和左室重量指数(LVMI=LVM/BW)。并取左室中段横截面心肌组织,用10%甲醛溶液(福尔马林)固定;采用采用胃酶酸解法测定心肌羟脯氨酸含量;采用饱和苦味酸天狼星红染色法测定大鼠心肌间质胶原表达的情况;采用免疫组织化学SABC法测定大鼠心肌局部AT1R、CTGF表达。

Objective: Sulfur dioxide(SO2) is considered to be one of the major components of atmospheric pollutants on the environment and harmful to human health。After being inhaled SO2 in the body fluid into its derivatives - sodium sulfite (Na2SO3) and sodium bisulfite (NaHSO3),through the blood of the body, which in other tissues and organs have an impact。Epidemiological study Studies show that exposure to SO2 gas can increase the long-term cardiovascular disease,such as arrhythmias, ischemic heart disease, pulmonary heart disease,etal. Mortality;SO2 in a dose-dependent inhibition of left ventricular pressure change rate,left ventricular pressure,heart rate and coronary 。The flow of heart function has negative inotropic effect。Long-term exposure to airborne fine particulate matter pollution in the environment,can cause fibroblast proliferation,collagen deposition,increased myocardial stiffness,reduced myocardial compliance。Studies have shown that aerobic exercise in rats。Myocardium to the positive development of myocardial interstitial components and components in real terms in proportion to growth and ultrastructure of a complete,Cxs increased and the normal configuration,play a good role of information flow and pulse transmission,is conducive to ventricular muscle contraction and relaxation of the overall consistency。Long-term .Adhere to the appropriate physical exercise or training,can lower blood pressure and improve heart function, increased cardiac reserve,reducing cardiovascular risk factors,improve the cardiovascular system function。SO2 pollution in the environment movement in the movement Will the myocardial interstitial collagen fibers in rats have not yet been reported. Laparotomy,thoracotomy the heart quickly removed,cut off the great vessels,ice salt water rinse,dry filter paper moisture,said the heart weight (HW) immediately after the saline in the ice along the atrioventricular junction of the rapid removal of left and right atria and large blood vessels,Removal of the right ventricular free wall,check the rest of the left ventricular mass (LVM), were calculated heart weight index (HBI) [HBI = heart weight (g) / body weight (g) X1000] and left ventricular mass index (LVMI = LVM / BW). And take the middle cross-section of left ventricular myocardium,with 10% formaldehyde solution (formalin) fixed;end of the experiment, measured by cardiac catheterization hemodynamics of rats in each group targets and calculate cardiac index;acid solution by stomach enzymes by determination of myocardial hydroxyproline content; using saturated picric acid Sirius red staining method for the determination interstitial myocardial collagen expression in the situation; by determination of immunohistochemistry SABC method for the determination of local myocardial AT1R,CTGF expression。Result: 1. Appropriate exercise can make the rat heart the morphological structure and function produce adaptability change,performance for cardiac weight, heart weight index,left ventricular weight index increased,myocardial appear adaptability hypertrophy;SO2 inhaled conditions, heart weight index,left ventricular weight index increased slightly;In SO2 inhalation and sports double stimulation condition,Ⅱ content and myocardial local Ang AT1R protein expression level increases can stimulate myocyte hypertrophy value-added, may cause a rat cardiomyopathy rational hypertrophy, performance for cardiac weight, heart weight index,left ventricular weight, left ventricular weight index significantly increased。2. Compared with the RG,EG group, SEG group heart weight, left ventricular mass, cardiac index, left ventricular index were significantly increased (P<0.05); SRG group left ventricular mass decreased significantly except (P <0.01),heart weight,cardiac index, left ventricular index were not statistically different. And compared with the EG,SEG group heart weight,left ventricular mass decreased significantly (P <0.01); cardiac index, left ventricular index SEG group compared with the EG group was not statistically different. 3. Compared with the RG, EG, SRG, SEG rat myocardial hydroxyproline content,collagen concentration increased significantly (P<0.01; P<0.05); and compared with the EG, SEG hydroxyl rat myocardium proline content,collagen concentration was significant difference (P<0.01); SRG compared with the EG group was not statistically different. 4. Compared with the RG,SRG,SEG myocardial interstitial collagen content of perivascular collagen content,CTGF expression was significantly increased (P<0.01; P<0.05);EG compared with the RG myocardial interstitial collagen content,vascular around the collagen content increased,but not statistically significant, and AT1R、CTGF expression was significantly decreased (P<0.01); compared with the EG,SEG myocardial interstitial collagen content of perivascular collagen content,AT1R、CTGF expression difference highly significant (P<0.01); the SRG compared with EG,perivascular collagen content,AT1R、CTGF expression significantly different ( P<0.01)。Conclusion: 1. Appropriate exercise can make myocardial small arteries increased perivascular collagen volume fraction, collagen fiber.Dimensional thickening;SO2 exposure in rat myocardium,collagen around the small arteries was significantly increased in the solvent fraction,especially under the conditions of SO2 exposure exercise myocardial small arteries around the excessive accumulation of collagen fibers。2. Appropriate exercise make rats myocardial,small arterial thickening collagen fiber around thickener,thick;Michigan Myocardial,small artery surrounding collagenous volume fraction increased slightly,myocardial collagen concentration significantly increased,myocardial collagen fiber happen to reshape;SO2 inhaled make rats thickening collagen fiber myocardial multiplied,arranging disorderly,coarse arteriole surrounding collagenous myocardial,volume fraction,myocardial collagen concentration significantly increased; SO2 inhaled and exercise double stimuli to rat myocardial collagen fiber and the myocardial protein composition produce more disadvantageous effects,myocardial collagen fiber bundle,mix a bulky into blocks accumulation,myocardial,small artery surrounding collagenous volume fraction, significantly more elevated cardiac collagen concentration。3. Appropriate exercise make rats in CTGF AT1R, myocardial tissue protein expression were significantly reduced; In SO2 inhalation and sports double stimulation condition,rats AT1R,CTGF myocardial local elevated levels of protein expression more significant。4.SO2 inhaled may through myocardial tissue AT1R,in effect CTGF protein expression level,which led to the rat myocardial collagen volume fraction,rise,AT1R myocardial collagen concentration,CTGF protein expression level of change may be SO2 influence myocardial collagen volume fraction,the main myocardial collagen concentration change mechanism。5. SO2 inhalation and exercise will not only offset movement double stimulation to the cardiovascular system beneficial health effects on cardiovascular system,and produce more unfavorable influences。Advice on body-building should pay attention to the sports environment selection, try to avoid the SO2 pollution sources。
