PET是核医学领域常用的临床检查影像技术。18F作为PET药物制备中最常用的放射性核素,具有射线能量适于成像、半衰期(T1/2 = 109.8 min)适于较复杂合成和远距离运输等优点;使用双功能连接剂络合[18F][AlF]2+与肽结合是一种新型的18F标记方法。 HBED-CC(N,N'-双[2-羟基-5-(羧乙基)-苄基]乙二胺-N,N'-二乙酸)是一种性能优良的双功能连接剂,广泛应用于68Ga,99mTc(CO)3,[18F]AlF等放射性核素的标记。关于68Ga标记HBED-CC-单侧和双侧取代衍生物研究结果表明HBED-CC-双侧取代标记物具有更优越的生物学性质。目前已有[18F]AlF标记HBED-CC-单侧取代衍生物的研究,标记和生物学结果都显示出较为优良的应用前景。本研究旨在建立[18F]AlF标记HBED-CC-双侧取代衍生物的新方法,并将单侧和双侧取代标记物的理化性质与生物学性质进行对比,拓广其应用范围,弥补研究空缺,拓展HBED-CC这一双功能连接剂的应用前景。 |

PET is a common clinical imaging technology in nuclear medicine. 18F has a suitable position energy for imaging, and a suitable half-life (t1/2 = 109.8 min) for radiosynthesis and long-distance transportation as a widely used radionuclide for PET imaging. Using bifunctional chelator (BFC) to chelate [18F][AlF]2+ with peptide is a new 18F labeling method. HBED-CC (N,N'-bis[2-hydroxy-5-(carboxyethyl)-benzyl]ethylenediamine-N, N'-diacetic acid), a bifunctional chelator with excellent properties, is widely used in the labeling of 68Ga,99mTc(CO)3,[18F]AlF and etc. The report of 68Ga labeled monomeric and dimeric HBED-CC derivatives showed that the dimeric HBED-CC compounds had better biological properties. At present, the report of [18F]AlF labeled monomeric HBED-CC derivatives showed excellent prospect in labeling and biological properties. The purpose of this study is to design a suitable method of labeling [18F]AlF with dimeric HBED-CC derivatives, and comparing the physical chemical and biological properties between monomeric and dimeric compounds. In order to expand the application of [18F]AlF, make up for the lack of research and expand the application prospect of HBED-CC. |
