RNAi是一种由一段外源或内源高度保守的双链RNA诱发同源mRNA高效特异降解,从而造成转录后水平的基因沉默现象。通过RNAi技术可以高效率、低成本地关闭特定基因的表达,RNAi被广泛应用于基因功能的探究。北京师范大学王猛将松材线虫脯氨酰4-羟化酶基因、衔接蛋白基因导入尖孢镰刀菌,饲喂松材线虫后发现松材线虫表现出不同程度的dumpy表型。本实验为王猛实验的后续实验,用王猛设计的转基因尖孢镰刀菌喂养松材线虫,观察干扰松材线虫脯氨酰4-羟化酶基因和衔接蛋白基因除了使松材线虫表现出dumpy性状外,是否产生其他性状;观察在转松材线虫衔接蛋白基因(Bx-apg-1)干扰松材线虫 7天、14天、21天后松材线虫的繁殖量、发育速度是否有变化。通过实验对线虫进行RNA干扰,结果为干扰松材线虫脯氨酰4-羟化酶基因和衔接蛋白基因对松材线虫除了dumpy表型外其他性状没有明显变化。用转衔接蛋白基因Bx-apg-1的尖孢镰刀菌喂养的松材线虫与用野生型尖孢镰刀菌喂养的线虫相比后代繁殖量更多,但是发育速率较慢。松材线虫对转入目的基因的尖孢镰刀菌取食量比野生型和转绿色荧光蛋白基因的尖孢镰刀菌多。

RNAi is the post-transcriptional gene silencing which is caused by a highly conserved exogenous or endogenous double-stranded RNA induced the homologous mRNA highly efficient and specific degradation. RNAi technology can close a specific gene expression with high efficiency and low cost, hence it is widely used in gene function exploration.Wang Meng, a Doctor of Beijing Normal University, introduced the prolyl 4-hydroxylase gene and the adaptin gene of the pine wood nematode into Fusarium oxysporum, and the pine wood nematodes showed different degrees of dumpy phenotype after RNAi. This experiment is the follow-up experiment of Wang Meng, using transgenic Fusarium oxysporum designed by Wang Meng to feed pine wood nematode, observed interference pine wood nematode prolyl 4-hydroxylase gene and adaptin gene of the pine wood nematode in addition to showed dumpy traits, but also do not produce other traits. Adaptin gene(Bx-apg-1) was used to observe the propagation and development of pine wood nematode after interference 7 days, 14days, and 21days. The results showed that the proline 4 - hydroxylase gene and the adaptin protein gene of Bursaphelenchus xylophilus did not change the other traits except for the dumpy phenotype. Compared with the nematode fed by wild-type nematode, the multiplication rate of the mutant protein gene Bx-apg-1 was more, and the development rate was slower.
