葡萄糖等营养物质刺激小鼠胰岛β细胞分泌胰岛素,其中钙离子是该刺激-分泌偶联的关键调控因子。葡萄糖通过胰岛β细胞质膜上的葡萄糖转运蛋白2进入细胞,在胞质中进行糖酵解生成丙酮酸、NADH及ATP;丙酮酸参与三羧酸循环生成更多ATP,升高胞内ATP/ADP比率,使ATP敏感性钾通道关闭,细胞膜去极化。细胞膜去极化导致电压门控钙通道的开放,胞外钙内流触发胰岛素分泌。胞内钙信号产生及调控途径,是研究胰岛素分泌调控的基础,也是探讨糖尿病病理及寻找治疗途径的基础。然而,现今对于单个小鼠胰岛β细胞胞浆钙振荡的研究甚少。本实验应用Fura-2荧光测胞浆钙浓度的方法,从细胞水平展现Rin-m5F细胞胞浆钙振荡所表现的特征。 |

Ca2+ plays an important role in glucose-stimulated insulin secretion in mouse β-cells. Glucose is transported into β-cell through GLUT2,and metabolized via glycolysis to produce Pyruvate, NADH and ATP.Pyruvate is further metabolized via the citric acid cycle to produce more ATP, which eievates the cytosolic ATP/ADP ratio. This raised ATP/ADP ratio induces closure of KATP in the plasma membrane,leads to plasma membrane depolarization and Ca2+influx through voltage-gated Ca2+ channels,to ultimately trigger insulin secretion. The mechanism of intracellar Ca2+ signal generation is fundamental for insulin secretion,and has important implications for the pathology and the treatment of diabetes. I therefore in this work investigated oscillations induced by glucose and other nutrients in single mouse β-cell-Rin-m5F cells. |
