本文主要研究了新型手性试剂 mco1 对高效毛细管电泳手性拆分喹诺酮类药物(氧氟沙星)的影响。新型手性试剂 mco1是新合成出来的一种手性位移试剂,其水溶性比较差。本研究通过加入有机溶剂对其助溶,手性试剂 mcol 可以在一定条件下拆分氧氟沙星对映体。同时,我们对 mco1 拆分氧氟沙星对映体的适宜条件进行了初步探究。 利用熔硅毛细管柱,在磷酸盐缓冲溶液中,研究了新型手性试剂 mcol对拆分氧氟沙星对映体的影响,初步探索的合适实验条件为磷酸盐缓冲溶液浓度为100mM,pH=4.15,合成手性试剂mco1浓度为 0.22mg/mL,样品浓度为:1mM。毛细管柱为50μm×54.6cm(有效柱长为41.8cm),进样时间为 3s,操作电压为15KV,紫外检测波长为254nm.我们具体研究了有机溶剂种类、缓冲溶液pH、不同缓冲体系对分离的影响。 |

The aim of this work was to study the effect of new chiral reagent on separating fluoroquinolone antimicrobial agents(ofloxacin) by means of high performance capillary electrophoresis.As a new chiral reagent that has just been synthesized, mcol has very low solubility in water.As been assistantly dissolved by organic solvent,mcol was found to be efficient for the enantioresolution of racemic ofloxacin at a certain condition. Further more, primary study on the feasible conditions for the ofloxacin enantioresolution was performed. In this work, with unfused capillary tube and 100mM phosphate(pH=4.15) as the buffer,and mcol(concentration:2.2x10-4g/mL)as an chiral additive to enhance the separation of ofloxacin enantiomers. We have studied the effects on the resolution, such as different organic solvents, pH and different buffer types and so on. |
