微丝骨架在许多细胞活动,如细胞分裂和分化、胞质环流和细胞形态建成中扮演重要角色,微丝骨架的动态变化是植物细胞许多生理活动的基础。微丝骨架的高度动态是在许多细胞因子的调控下完成的,微丝骨架结合蛋白就是其中一类重要的调节因子,近几年来该领域的研究相当活跃。本研究中涉及的Fimbrin蛋白就是一种分子量为68-75kDa的肌动蛋白成束蛋白,它被作为动物细胞微绒毛和其他细胞表面结构中微丝成束的主要结构蛋白被发现。目前对Fimbrin的了解主要集中在动物和酵母中,而关于植物fimbrin蛋白的功能和生化特性的研究结果还非常有限。
我们实验室从百合中得到fimbrin的一种同源蛋白,命名为Lfimbrin,并构建了Lfimbrin的真核表达载体1300-Lfim-GFP。通过基因枪法将其转入洋葱内表皮细胞中,期望通过其过量表达,以研究fimbrin的体内功能及亚细胞定位。结果显示对照组GPF在细胞质及细胞核分布;与对照组相比较,实验组Lfim-GFP在细胞质中有荧光,而细胞核内无。荧光定位研究的初步结果表明,fimbrin可能定位在细胞质中。

The actin cytoskeleton plays a key role in many cellular processes including cell division and differentiation, cytoplasmic streaming, and in governing plant-cell morphogenesis. The dynamic change of actin cytoskeleton is the base of plants′ physiological activities, which are regulated by many cell genes. The actin cytoskeleton bounding proteins are one of the most important regulating genes. In recent years, the study of this area is very active. Fimbrin protein referring in this study is an actin whose molecular weight is 68-75kDa. It was found as main structural proteins in animal cell floss and in other cell exterior structures. Until now, the cognition of Fimbrin mainly focus on animals and yeast, however, The study of the function and properties of fimbrin proteins from plant cell is quite limited.
We got one of fimbrin′s homologous genes from Lilium, naming Lfimbrin. We also construct carrier 1300-Lfim-GFP, which is transmitted into onion inner epidermal cells by gene gun. To visualize the subcellular localization of Lfimbrin, we have over-expressed Lfim-GFP in Allium. The result is that GPF is distributed in cytoplasm and nucleus. By contrast, Lfimbrin is only distributed in cytoplasm. The preliminary results show that fimbrin may be targeted in the cytoplasm.
