Aβ斑块在大脑中沉积是老年性痴呆症的主要特征, Aβ斑块可作为诊断老年性痴呆症的生物标志物, 研发特异性的Aβ斑块显像剂对老年性痴呆症的诊断和监测是非常重要的。本文用荧光光谱法对一系列黄酮类化合物与Aβ斑块的亲和力进行了定性分析,从中发现有四种黄酮化合物与Aβ斑块有较高的亲和力,这些黄酮化合物经修饰后可以用放射性核素18F或者99mTc去标记。同时,还合成了两种新的黄酮化合物:6-甲氧基-4'-胺基黄酮和6-甲氧基-4'-N,N二甲基胺黄酮,和FDDNP的前体化合物。另外,以[NO][BF4]和[99mTc(CO)3(EHIDA)]-为原料分别在乙腈和水中成功制备了[99mTc(CO)2(NO)( EHIDA)]0,其产率可达99%以上,并用TLC、HPLC和电泳等分析手段验证了实验结果,还初步探讨了[99mTc(CO)2(NO)(EHIDA)]0在昆明鼠体内的生物分布,为在水中制备高产率的[99mTc(CO)2(NO)]2+核提供了一种简易方法。

The accumulation of Aβplaques in the brain is a key factor associated with Alzheimer’s disease (AD). Aβplaques in the brain may be useful as a biomarker for diagnosis of AD. Development of Aβplaque-specific imaging agents is important for the diagnosis and monitoring of AD patients. The affinity of a series of flavone compounds for Aβplaques has been assayed by fluorescence binding determination, and four flavone compounds showed high binding affinity for Aβplaques. These compounds can be labeled with 18F or 99mTc after have being modified. Two new flavone compound: 6-methoxy-4'-aminoflavone and 6-methoxy-4'-dimethylaminoflavone, and 2-(1,1-dicyanopropen-2-yl)-6-2-hydroxyethyl)-methylamino)-naphthalene have been synthesized. A novel compound [99mTc(CO)2(NO)(EHIDA)]0 has been prepared by reacting [99mTc(CO)3(EHIDA)]– with [NO][BF4] both in water and acetonitrile. The conversion of [99mTc(CO)3(EHIDA)]– to [99mTc(CO)2(NO)(EHIDA)]0 was supported by TLC, HPLC and eletrophoresis. The radiochemical purity (more than 99%) was proved by TLC and HPLC. The biodistribution in mice was also studied.
