贵金属纳米团簇(Noble Metallic Nano Clusters,NMNCs)通常是指由Au、Ag或Pt等贵金属的几个至几十个原子组成的具荧光、水溶性的分子级聚集体。其特有的量子尺寸效应使其光学性质具有随粒径大小而变化的特性,这种特性使其荧光发射光谱在可见光到近红外光区范围内可调谐。Au、Ag或Pt等金属具有化学惰性,其保护基团对生物体的毒副作用小,使得贵金属纳米簇具有良好的生物相容性。近几年来,蛋白质由于其温和的反应条件、尺寸控制、高度特异性选择及多功能性,常作为合成贵金属纳米簇的模板。各种各样的贵金属纳米簇都成功地以蛋白质为模板获得。本文使用溶菌酶为模板在碱性条件下合成银纳米簇,并对其光学性质进行检测和分析。将合成的银纳米簇和与溶菌酶特异性识别的DNA在一定条件下反应,探究其荧光性质的改变,研究其机理。并提出了对于以溶菌酶为模板的银纳米簇在未来生物医学领域及日常生活中的应用的展望。
外文摘要:
Noble metal nanoclusters (NMNCs) are the molecular aggregates with fluorescence and water-soluble, which are composed of several to dozens of atoms, such as Au, Ag, and Pt atoms. Its unique quantum size effect makes the optical properties has the character of changing with particle size, and this feature makes the fluorescence emission spectra within the scope of visible and near infrared region become tunable. Au, Ag and Pt metal has chemical inertness, the side effects of its protecting groups to organisms is small, which causes precious metals nano clusters has good biocompatibility. In recent years, proteins due to its mild reaction conditions, choice of size control, highly specific and versatility, often used as a template in the synthesis of noble metal nano clusters. All kinds of precious metals nano clusters are successfully compounded with protein as template. This article using lysozyme as the template to compound silver nanoparticles cluster in alkaline conditions, and the optical properties is tested and analysed. The silver nano clusters and the DNA which has specific recognition to lysozyme react under certain condition, to explore the change of the fluorescence properties and to explore its mechanism. And this article puts forward the application prospect of the silver nano clusters with lysozyme as a template in biomedical field and of daily life in the future.
