Hsp90是热休克蛋白家族的一员,具有高度保守性和普遍性。该蛋白是真核细胞中含量最多的蛋白质之一,主要参与细胞内蛋白质的折叠、转运、活化、降解及蛋白信号的呈递,是生物的基本生命活动所必需的。果蝇中Hsp90的同源物为Hsp83。已有研究显示Hsp83与生物钟核心调控基因相关(per,tim,cyc,clk)。课题组前期实验还筛选得到四个受到Hsp83影响的基因,包括三个参与生物钟次级调控通路的基因(pdp1,vri,cwo),以及睡眠关键基因wake。暗示着Hsp83不仅可能参与节律基因的调控,还可能会对果蝇的睡眠产生影响。本研究对中枢神经系统过表达Hsp83的果蝇(Hsp83Elav O/E)的睡眠稳态调节能力进行探究。研究结果显示,在正常情况和轻度睡眠压力的情况下,Hsp83Elav O/E果蝇的睡眠质量不如野生型果蝇(w1118);但在长期睡眠剥夺时,Hsp83Elav O/E会立即调整自身睡眠模式,例如延长睡眠时间、改善自身“睡眠颠倒”现象,进而降低极端条件下的死亡率。这说明增加Hsp83的表达确实会对果蝇的睡眠产生正向影响,尤其在极端条件下能显示其优越性。实验结果显示,w1118果蝇中Hsp83的表达具有节律性,但Hsp83Elav O/E果蝇中Hsp83的节律性被破坏,这可能是Hsp83Elav O/E果蝇“睡眠颠倒”的原因。总之,实验结果显示过表达Hsp83的果蝇会在极端条件下显示出优越性,死亡率更低。因此,为了能在正常情况下也产生正向作用,Hsp83过表达的模式还需调整,希望能按照其自身的节律性同步放大。

As a member of the heat shock protein family,Hsp90 is highly conserved and universal,and is one of the most abundant proteins in eukaryotic cells.It is involved in the folding,transport,activation,degradation and presentation of protein signals in cells,and is essential for the basic life activities of organisms.The homologue of Hsp90 in Drosophila melanogaster is Hsp83.Previous studies have shown that Hsp83 is related to the core regulatory genes of circadian clock(per,tim,cyc,clk).Our previous experiments also screened four genes affected by Hsp83,including three genes involved in secondary circadian clock regulation pathways(pdp1,vri,cwo),and the key sleep gene wake.This suggests that Hsp83 may not only be involved in the regulation of rhythm genes,but also affect the sleep of Drosophila melanogaster.This study investigated the sleep homeostasis regulation in Drosophila with central nervous system overexpression of Hsp83(Hsp83Elav O/E).The results showed that the sleep quality of Hsp83Elav O/E was inferior to that of wild-type Drosophila(w1118)under normal conditions and mild sleep pressure.But in the case of chronic sleep deprivation,Hsp83Elav O/E immediately adjusts its sleep pattern,for example by extending sleep duration and improving its sleep-reversal phenomenon,thereby reducing mortality in extreme conditions.This suggests that increasing the expression of Hsp83 does have a positive effect on Drosophila sleep,especially under extreme conditions.The results showed that the expression of Hsp83 in w1118 was rhythmical,but the rhythmical expression of Hsp83Elav O/E was destroyed,which may be the cause of sleep reversal in Hsp83Elav O/E.In conclusion,the results show that flies overexpressing Hsp83 show superiority under extreme conditions with lower mortality.Therefore,in order to have a positive effect under normal conditions,the overexpression pattern of Hsp83 needs to be adjusted,hoping that it can be amplified synchronously according to its own rhythm.
