【目的】利用基因芯片研究石杉碱甲对老年性痴呆模型大鼠基因表达的影响。【方法】于20只雄性SD大鼠海马区注射Aβ25-35,将其随机分为药物组与模型组。造模24h后,药物组大鼠进行石杉碱甲灌胃处理30天。对脑组织中提取物进行基因芯片技术相关操作,获取药物组与模型组相关基因表达水平的差异,评价石杉碱甲对老年性痴呆的药物疗效。【结果】在石杉碱甲的作用下,老年性痴呆模型大鼠细胞中表达水平上调的基因有110个,下调基因有150个,这些基因的功能涉及转录调节、细胞信号转导,离子通道,免疫反应、基础代谢等。【结论】石杉碱甲对老年性痴呆有明显疗效,实验结果对研究石杉碱甲对抗老年性痴呆的药物靶点及其副作用可能有帮助。

【Objective】Using cDNA microarray to study the effect of Huperzine A on the gene expression of AD rat models.【Methods】Injecting Aβ25-35 in the hippocampus region of 20 male SD rats to form the AD rat models. After 24 hours, the rats were divided into two groups. One was treated by Huperzine for 30 days, and the other as control. Fluorescence-labeled cRNA probes generated from AD rat model were hybridized with a cDNA microarray. By comparing the data between the groups, the effect of Huperzine A on the expression of associated genes were evaluated.【Result】110 up- regulated genes and 150 down-regulated ones were screened out by the comparison of the expression level. Those genes encode proteins involved in regulation of transcription, signal transduction, ion channel, immune response, basic metabolism and so on.【Conclusion】Huperzine A was highly efficient in AD. The result may be helpful in studying its targets and side effect.
