建立可靠的多级别的胶质瘤动物模型对于进一步了解和研究胶质瘤发病 机制及治疗方法有重要意义。目前人脑胶质瘤的异种移植动物模型常采用不 同免疫背景的大鼠和小鼠模型,其中高级别的胶质瘤模型比较成熟,但低级 别的胶质瘤模型成功率不高。造模时,移植脑区通常为纹状体,由此想到鸣 禽纹状体终生招募新生细胞,特别在出生后幼年阶段,有大量神经细胞的加 入,而小鼠纹状体的新生细胞则很少。因此,推测鸣禽纹状体的微环境很可 能更有利于新细胞的存活和发育,为在小鼠中较难建立的低级别胶质瘤模型 提供可能。本课题利用脑立体定位技术,将荧光标记的 U-87MG 人脑星形胶质 母细胞瘤细胞注入幼年鸣禽斑胸草雀的纹状体内,因为这个肿瘤细胞系恶性 程度较高,在大小鼠异种移植中移植常用且较成熟,以提高在鸣禽中实验的 成功率。脑内注射后进行肌肉注射免疫抑制剂,抑制免疫排斥反应,通过灌 流、切片检测肿瘤的生长,结果显示部分实验动物脑内可能有异种组织形成, 可能为进一步探索人脑胶质瘤鸣禽脑内移植模型的构建提供支持,但仍需进 一步利用免疫组织化学等技术进行鉴定。
外文摘要:
The establishment of a reliable multi-level glioma animal model is of great significance for further understanding and research on the pathogenesis and treatment of glioma. At present, the xenograft animal models of human glioma often use rat and mouse models with different immune backgrounds. Among them, high-grade glioma models are relatively mature, but low-grade glioma models have a lower success rate. When modeling, the transplanted brain area is usually the striatum, so it is thought that the songbird striatum recruits new cells throughout its life, especially in the early postnatal period. But in mice, it is fewer. Therefore, it is speculated that the microenvironment of the songbird striatum is likely to be more conducive to the survival and development of new cells, providing a possibility for low-grade glioma models that are difficult to establish in mice. This project used the brain stereotaxic technology to inject fluorescently labeled glioma cells into the striatum of the juvenile songbird zebra finch, and intramuscularly inject immunosuppressive agents to suppress immune rejection. The growth of tumors is detected by perfusion and slices. It was shown that tumors formed in the brains of some experimental animals, which provided support for further exploration of the construction of human glioma songbird brain transplantation models.
