驱动蛋白是一种重要的生物分子马达,它广泛存在于真核细胞表面和真核细胞内,可以利用水解三磷酸腺苷(ATP)产生的化学能,转化为机械能,从而定向输运胞内各种物质来保证生物体的正常生命活动。本文首先调研了驱动马达的生物学背景、相关实验结果以及理论模型的研究进展,以非平衡态随机动力学方程为基础构造了描述驱动蛋白定向运动的理想模型;接着修正了理想模型中有效自由能的值,并考虑酶促反应中活化能势垒E的影响,引入一个包含较少参量却能反映驱动马达在行走中机械化学耦合的非对称周期势,研究了驱动蛋白在非对称周期势中的随机动力学行为,由此模型推导出的平均速度、失速力、随机性等值均可表示为ATP浓度[ATP]和外部荷载F的函数;最后用Matlab分别对理想模型和改进模型进行数值模拟,将模拟所得图像与实验记录所得图像进行分析对比,发现理想模型在一定程度上能体现驱动蛋白运动的定向性,但图像细节处与实验图像相差较大,而改进模型所得结果与实验图像基本吻合,从而验证了模型的合理性。 |

Kinesin is an important biomolecule motor, which widely exists on the surface and inside of eukaryotic cells, and ensures the normal life activities of organisms by directionally transporting various intracellular substances. In this paper, firstly, we investigate the biological background,related experimental results and research progress of theoretical model of kinesin,constructing an ideal model which is based on the non-equilibrium stochastic dynamics equation to describe the directional motion of kinesin. Then, by modifying the value of effective free energy in the ideal model and considering the effect of activation energy barrier E in enzymatic reaction , we introduce an asymmetric periodic potential which contains few parameters but can reflect the chemical-mechanical coupling of the kinesin, and study the random dynamic behavior of kinesin in the asymmetric periodic potential. The average speed, stall force and randomness obtained from this model can be expressed as a function of ATP concentration and external load F. Finally, we numerically simulate the models by Matlab, comparing and analyzing the simulated images with the experimental images. It is found that the ideal model can reflect the directionality of kinesin movement, but the details are quite different from the experimental images, but the results of the improved model are basically consistent with the experimental images, which verifying the rationality of the model. |
