前列腺的发展过程涉及繁多信号通路和蛋白,是一个非常复杂的过程。这其中,尤其是原位肿瘤迁移的过程中,胞外基质蛋白发挥着重要的作用。在前期研究中,本实验室使用 ICAT 技术分析的分泌蛋白中,CYR61( Cysteine-rich,angiogenic inducer, 61; Insulin-1ike growth factor-binding protein 10; ProteinGIG1)是一个重要成员。研究已发现,CYR61 能够促进细胞增殖、趋向发展、新血管形成,和细胞粘连。在多种癌症中均发挥作用。 为了进一步研究 CYR61 在前列腺癌中的表达差异及作用,本实验首先运用 RT-PCR验证了蛋白质组学的结果,另外,我们运用 Western blot 和免疫组化手段同样证明了CYR61 在原位前列腺癌和转移前列腺癌组织中与正常组织存在表达差异。在此基础上,我们构建了 CYR61 的过表达和沉默质粒,从多个角度分析CYR61在前列腺癌的转移发展过程中所发挥的作用。本论文研究为进一步开展CYR61 的功能奠定了一定基础。 |

A great amount of signal pathways and proteins are involved in the development of prostate cancer, which is, maybe, the most difficult process to understand by now. In this process, especially when the metastasis of tumor in situ happened, proteins in the extracellular matrix play a significant role. In the former research, ICAT analysis was used to detect secretion protein and found out a promising member, CYR61. So far as we know, CYR61 is related to various cell activities such as proliferation, migration, Angiogenesis, cell adhesion, and exerts its influence in many cancers. In order to find the different exsspression and role of CYR61 in prostate cancer, Members in our lab validated the results from ICAT using RT-PCR.I affirmed the results by western blotting and immunohistochemistry. Following these data, we constructed a series of plasmids including those used to overexpress and silence CYR61 in cell lines in order to analyze its function in metastasis. The research in this paper will give some basic advice to deepresearching for CYR61 function analysis. |
