球毛壳菌(Chaetomium globosum)广泛分布于自然界,可以降解木质纤维素,合成大量具有生物学活性的次级代谢产物,在生物制药和生物防治中具有重要作用。其最重要的次级代谢产物球毛壳菌素A(Chaetoglobosin A,ChA)属于细胞松弛素类,具有抗真菌、抗癌、抗线虫活性,可开发为抗癌药物,生物农药。因此提高ChA产量,研究其生物合成的调节路径成为近年来的研究热点。本研究利用基因缺失的方法探究全局性调控因子MADS-box转录因子在球毛壳菌中的功能。首先以构巢曲霉(Aspergillus nidulans)MADS-box转录因子氨基酸序列为参考,在球毛壳菌基因组中找到同源基因:CHGG_01378和CHGG_06470。通过蛋白序列比对,确认表达量较高的CHGG_06470基因属于MADS-box蛋白家族中的SRF型MADS-box转录因子基因。再结合CRISPR-Cas9技术构建表达量较高的CHGG_06470基因McmA的敲除质粒pKMCMA。原生质体转化之后得到56株具有标记基因潮霉素抗性的转化子,最后得到4株McmA基因缺陷菌株。缺陷菌株的产孢能力均受到影响,初步判断MADS-box转录因子McmA影响球毛壳菌的产孢能力。

As an important secondary metabolite of Chaetomium globosum, chaetoglobosin A,ChA a has anti-fungal, anti-cancer and anti-nematode activities, and plays an important role in biopharmaceutical and biological control.The global regulatory factor MADS-box transcription factor plays an important role in the growth, development and reproduction of fungi. We study the function of MADS-box transcription factor in C. globosum by gene deletion. Firstly, homologous genes, CHGG_01378 and CHGG_06470, were found in the genome of C. globosum by reference to the amino acid sequence of the MADS-box transcription factor of aspergillus structurus. Through protein sequence alignment, it was confirmed that the CHGG_06470 gene with high expression level belongs to the SRF-like MADS-box transcription factor in the MADS-box protein family.The knockout plasmid pKMCMA of CHGG_06470 gene McmA was constructed by combining with CRISPR-Cas9 technology. After protoplast transformation, 56 transformants with hypomycin resistance were obtained, and finally 4 strains with McmA gene defect were obtained.The spore-producing ability of the defective strain was weakened and the colony growth rate was slow.It was preliminarily determined that MADS-box transcription factor MCMA affected the spore-producing ability and growth and development of C. globosum.
