肠道是消化系统重要的组成器官之一,在营养物质消化和吸收、微生物免疫及激素调节中发挥了重要作用,但肠道具体是如何通过各细胞类型的分工及互作调控实现其生理功能的目前仍认识不足。近些年来已经有一些针对正常肠道的单细胞转录组研究,然而转录水平仅能体现基因表达的中间状态,并不能完全反映细胞功能的执行体——蛋白质的水平,因此蛋白质组较转录组能更为精准地反映细胞、组织及器官如何维持正常生理功能。本课题最终目标为开展细胞类型分辨率下的肠道蛋白质组研究,本论文完成了要实现该目的所需的前期工作,即建立并优化了肠道上皮层单细胞悬液制备方法,并利用肠道单细胞转录组数据及膜表面蛋白数据库分析确定可用于分离出肠道上皮层各类细胞类型所需的流式分选标志物。 |

As one of the significant components of the digestive system, intestine plays an important role in nutrients digestion and absorption, microbiome defense and hormone homeostasis and regulation. However, it remains largely unknown how intestine achieves its functions through the division of labor and cross-talk among various cell types. In the past few years, there are some existing literature focusing on single-cell transcriptome of small intestine, but the level of transcription just reveals the level of mediator, not the final product of gene expression, protein, which is the executor of functions of cells. Thus, proteome has advantages over transcriptome of reflecting how cells, tissues and organs maintain their physical functions. To build a cell-type-resolved adult mouse intestine proteome eventually, this paper provides preparation for the above goal. The optimized protocol for obtaining mouse intestinal epithelium single cell suspension is established, and the specific markers of various cell types in intestinal epithelium for fluorescence-activated cell sorting are screened out through analysis of scRNA-seq data and database of subcellular locations of human proteins. |
