可降解烟碱的微生物在烟草工业中具有重要的应用价值。马铃薯块茎蛾幼虫能取食烟叶,其肠道微生物中可能具有降解烟碱的微生物。本论文通过将马铃薯块茎蛾幼虫肠道细菌的烟碱培养基平板分离和16SrDNA序列测定,共筛选和鉴定出对烟碱具有耐受作用的细菌共20种,隶属于4个门,7个目,14个属,其中变形菌门和厚壁菌门占比较高。应用相关软件(MEGA 6)构建了这些细菌的系统发育树。分离的细菌经烟碱选择性培养纯化后,接入40ml体系烟碱培养液中培养24小时,按0h、2h、4h、 8h、12h、24h时间点测定菌液浓度(OD值)随时间的变化,结果显示,蜡样芽胞杆菌(Bacillus cereus)、无乳链球菌(Streptococcus agalactiae)、乙酸钙不动杆菌(Acinetobacter calcoaceticus)、阿氏肠杆菌(Enterobacter asburiae)等四种细菌生长幅度较其他菌株明显,而湖南鞘氨醇单胞菌(Sphingomonas hunanensis)、鲁氏不动杆菌(Acinetobacter lwoffii)、红球菌属(Rhodococcus cerastii)、蒙氏肠球菌(Enterococcus mundtii)、戴尔福特菌(Delftia tsuruhatensis)、氧化微杆菌(Microbacterium oxydans)等六种细菌生长呈上升趋势,其余十种菌株生长曲线较平缓,表明它们利用烟碱作为唯一碳源的能力有明显不同,利用烟碱能力强的细菌具有开发和应用潜能。

Microorganisms that degrade nicotine have important application value in the tobacco industry. The Phthorimaea operculella can feed on tobacco leaves, and their intestinal microorganisms may have ability to degrade nicotine. In this study, 20 bacteriathose may tolerable of nicotine were isolated from the intestines of Phthorimaea operculella larvae by culture on nicotine plates and identified by 16SrDNA sequences, which belong to 14 genera 7 orders 4 phyla. Of which, Proteobacteria and Firmicutes have a relatively high proportion. The phylogenetic tree of these bacteria was constructed by using the software MEGA 6. After purification culture, each strain was cultured in 40 ml of nicotine liquid medium for 24 hours. The concentration of bacterium (OD value) was measured at time points of 0h, 2h, 4h, 8h, 12h and 24h. The result showed that, the growths of Bacillus cereus, Streptococcus agalactiae, Acinetobacter calcoaceticus and Enterobacter asburiae were more obvious than that of other strains; while, the growths of other six bacteria including Sphingomonas hunanensis, Acinetobacter lwoffii, Rhodococcus cerastii, Enterococcus mundtii, Delfitia tsuruhatensis and Microbacterium oxydans were also somewhat increased; however, the remaining ten strains are almost not increased, indicating that their abilities to utilize nicotine as the sole carbon source are significantly different. The bacteria with strong ability to degrdating nicotine have a wide prospect for development and application.
