本文建立了介孔材料基质辅助聚丙烯酰胺凝胶电泳分离血清蛋白质的新方法。有序介孔材料SBA-15经聚乙二醇处理后均匀分散在聚丙烯酰胺凝胶的溶液中,分别加在一维凝胶的浓缩胶和分离胶以及二维凝胶中,利用SBA-15的不同作用来辅助聚丙烯酰胺凝胶电泳分离血清蛋白质。SBA-15在浓缩胶中对高丰度蛋白质如免疫球蛋白G(IgG)的吸附作用提高了低丰度蛋白质的分辨率;SBA-15在分离胶中对低分子量蛋白质的富集作用,使一些低分子量蛋白质如维他命D结合蛋白等经过传统的考马斯亮蓝染色后可以在胶上清晰地观察到其条带。同时,研究了在胶的相同位置加入无序介孔材料硅酸钙(CS),发现在分离效果方面SBA-15优于CS。SBA-15和蛋白质之间结合力的类型有范德华力、氢键等,通过结合力使蛋白质被吸附,但由于它们之间的结合力比较弱,在电泳的条件下这些蛋白质又会被解吸附,从而和其他蛋白质分离开。SBA-15通过调节蛋白质的电泳速率辅助聚丙烯酰胺凝胶电泳分离血清蛋白质,这一方法开阔了介孔材料在分离蛋白质方面应用的新领域。

In this thesis, a novel application of ordered mesoporous SBA-15 for improving the resolution of a native polyacrylamide gel electrophoresis (PAGE) in the detection of serum proteins is described. SBA-15 is dispersed homogeneously by polyethylene glycol (PEG) in the gel solution and has been respectively introduced into 1-D and 2-D PAGE system when the gel is manufactured. SBA-15 plays different roles when it is added to different regions in assisting PAGE separating serum proteins. In the stacking gel, it adsorbs high abundant proteins such as immunoglobulin G (IgG) thus resolution of low abundant proteins is improved. In the separating gel, it plays the role of enrichment for low molecular weight proteins (LMWP) through adsorption and then desorption to regulate the electrophoretic mobility, as a result some LMWP such as Vitamin D-binding protein are observed clearly in the gel after CBB-R250 detection. Comparing to disordered mesoporous material calcium silicate (CS), SBA-15 is better in separating. It adsorbs proteins through interaction with proteins such as van der waals force, hydrogen bond and then desorbs some due to weak interaction. SBA-15 through regulating electrophoretic mobility of proteins assisted PAGE separating serum proteins, which expands novel application of mesoporous material in the area of separating proteins.
