随着人类生活水平的提高,肥胖已成为备受关注的一个公众话题,是当今社会人们面临的重大难题之一。近年来,与肥胖密切相关的脂质代谢及其相关研究成为当今生命科学研究领域的热点之一。
本实验室一直致力于脂滴生物学的相关研究。脂滴生物学的研究对于肥胖以及多种代谢类疾病可谓息息相关,甚至是突破点之一。近年来,越来越多的研究着重于脂滴的形成和调控。本项目将着眼点放在了LRP1对于脂质,尤其是中性脂储存的影响上。
利用脂滴的分离纯化技术得到了脂滴。通过对脂滴进行相关生物化学实验和分析,初步证实了LRP1确实对于脂质储存,即脂滴有一定的影响。从形态学上,通过Nile Red染色,观察到LRP1基因缺失(LRP1-/-)会导致细胞内脂滴变小。并初步获得了脂滴的生物化学相关概况。

With the improvement of human beings living standards, obesity has become a public topic of great concern, which is one of the major problems faced with people in modern society. Lipid metabolism as well as its related researched, tightly associating with obesity, has emerged to be one of the very hotspots among the life sciences fields recent years.
Our laboratory has been committed to biological research of lipid droplets. Research of lipid droplets biology is closely binding to obesity and various metabolic diseases. More of, it is the key point of this research area. There is an increasing number of studies that concentrates on the biogenesis and regulation of lipid droplets. In this study, focus was put on the impact of LRP1 on neutral lipids, especially the impact on the storage of triglycerides.
Take advance of the techniques of lipid droplets isolation and purification, the purified lipid droplets were obtained. Through the related biochemical experiments and analysis, the impact of LRP1 on lipids storage was identified, which means LRP1 can influence the lipid droplets. Morphologically, the size of the lipid droplets in LRP1 (LRP1-/-) deficient cells becoming smaller was observed using Nile Red staining. The related biochemical profile of the lipid droplets also was explored.
