脆杆突变的研究,以往主要集中在遗传分析,确定突变的显隐性关系,突变基因的数目和相关基因的染色体定位。迄今为止,水稻中报道的脆杆基因有6个,分别定位在染色体3(bc — 1),染色体 5(bc-2)染色体 2(be - 3,bc 一 5)和染色体 6(bc — 4). 其中,bc-5是茎节脆性基因,Bc 一6 是个脆杆显性基因,但是,尚未染色体定位。本 实验做的就是Bc一6的基因定位 本研究以水稻脆杆突变体为材料,通过形态学和化学方法详细对突变体进行鉴定,用细胞手段对细胞壁结构进行分析;再利用遗传学的方法与另一个具多态的野生型亲本 进行杂交,获得F1,自交配制 F2 群体;通过重组连锁理论,借助图位克隆方法确定控制基因的性质(显、隐性)和定位到特定的染色体区段,为最终克隆基因奠定基础。目前由于时间关系,实验只不过做到初定位。 |

The Brittle culm mutant of research, former main concentrate Be inherit analysis,assurance mutate of show the recessiveness relation, the mutation number of the gene and the chromosome of the related gene orientation.Up to the present, in the paddy rice the mutant gene of the report way have 6, positioning in the chromosome respectively 3(bc-1),chromosome 5(bc-2), chromosome 2(be a 3, bc a 5) with chromosome 6.(bc a 4) Among them,the bc-5 is the caulis stanza brittleness gene, Bc a 6 is a frailty pole to show sex gene, but, the chromosome didn'toyet fixed position.What this experiment do is Bc-6 gene orientation This research takes the Brittle culm mutant rice as material, learning through an appearance with the chemistry method to carry on authenticate towards mutating a body in detail, using the cell means to carry on analysis to the cell wall structure;The method and another that make use of genetics again have a close origin of the wild type of Polymorphism to carry on miscellaneous hand over, acquire F1, from the mating system F2 colonys;Via the reorganization catena theories, ask for Map-based cloning to ascertain the quality(show,recessiveness) of the clone method assurance control gene with position to the particular chromosome block, lay foundation for the end clone gene. |
