Hsc70-4是热休克蛋白Hsp70家族的一员,在进化过程中具有高度保守性,在生物体发生保护性热休克应答过程中发挥着重要的作用。睡眠由节律系统和睡眠稳态共同调节,目前研究发现,Hsp70家族中的BiP/GRP78在维持果蝇正常的睡眠稳态中发挥关键作用。本实验利用实时荧光定量PCR技术和果蝇睡眠监测仪器针对Hsc70-4是否会影响果蝇的睡眠以及参与调控过程的分子机制进行初步探究。由于睡眠是由生物钟和稳态两个方面进行调控的,要探究Hsc70-4对睡眠稳态的影响,则需要探究Hsc70-4对节律的影响,因此,本次实验将通过实验研究Hsc70-4在中枢神经系统过表达或敲低后是否会影响节律调控核心基因,寻找相互作用的具体位点,以确定Hsc70-4在睡眠调控网络中的位置。本次研究发现,果蝇中枢神经系统过表达Hsc70-4后,可能对果蝇部分生物钟核心基因的震荡造成影响。

Hsc70-4 is a highly conserved member of the heat shock protein Hsp70 family, playing a critical role in the protective heat shock response in organisms during evolution. Sleep is regulated by both the circadian system and sleep homeostasis, and recent research has revealed that BiP/GRP78, a member of the Hsp70 family, plays a key role in maintaining normal sleep homeostasis in Drosophila. In this study, we used real-time quantitative PCR and Drosophila sleep monitoring equipment to preliminarily explore whether Hsc70-4 affects Drosophila sleep and the molecular mechanisms involved in its regulation. Since sleep is controlled by both the biological clock and homeostasis, to investigate the impact of Hsc70-4 on sleep homeostasis, it is necessary to explore the effect of Hsc70-4 on circadian rhythms. Thus, this study aimed to investigate whether overexpression or knockdown of Hsc70-4 in the central nervous system affects core circadian regulatory genes, and to identify specific interaction sites, to determine the position of Hsc70-4 in the sleep regulatory network. Our research findings suggest that overexpression of Hsc70-4 in the central nervous system of Drosophila may impact the oscillation of certain core circadian genes.
