Ca2+作为植物细胞内重要的第二信使,在维持细胞内稳态和植物生长发育过程中发挥重要的作用,是连接外界信号及环境胁迫和细胞生理生化反应之间的枢纽。目前已经发现多种刺激因素,如机械刺激、低温、红光、植物激素、真菌激发子、缺氧和水分胁迫等,作用于不同的植物细胞,最初反应几乎都是首先导致胞质内Ca2+浓度发生变化,并引起一系列的下游反应。同胞质钙信号的研究相比,植物细胞核内钙信号的研究甚少,对于植物细胞核内钙信号是否直接受到外界信号的调控还没有系统的认识。但大豆根瘤菌根结因子使植物细胞核内产生钙离子-钙调素结合蛋白激酶(CCaMK)的研究为分离受核钙信号调控的感知因子提供了重要的支撑。本研究项目以测定拟南芥细胞核内钙离子浓度的变化为手段,在整体水平系统地研究细胞核钙信号的发生形式及调控机制。将pFGC-NLS-Cameleon YC3.6表达载体转入拟南芥后,成功获得了YC3.6核定位表达的转基因植株,并通过连续三代的种植筛选获得纯合种子。用植物激素(JA、ABA)和环境胁迫(高渗)等处理幼苗根尖细胞,利用LSM激光共聚焦显微技术实时监测细胞核内钙离子浓度的变化。
外文摘要:
As an important second messenger, Ca2+ plays a key role in maintaining cell homeostasis and regulating plant growth and development. It has been reported that many stimulus factors such as mechanical stress, low temperature, red light, plant hormone, fungal elicitor, hypoxia and water stress, can cause the changing of cytosolic calcium level and a series downstream reactions. However, little research on nuclear calcium signaling is processed systematically, and whether nuclear calcium signal is regulated directly by outer signal is still unknown. But that the soybean rhizobia nodular factor causes the formation of Calcium- Calmodulin dependent protein kinase (CCaMK) supports the research of isolating the Calcium related factor regulated nuclear calcium signaling. In this study we try to reveal the mechanism of nuclear calcium signaling and its regulation by measuring the nuclear calcium level. With constructing the express vector pFGC-NLS-YC3.6 and transferring it into Arabidopsis, we got the homozygous seeds which can express YC3.6 protein by three generation’s screening. The material was Arabidopsis seedling root tip cell, treated with hormone (JA, ABA) or environment stress (high osmosis) and the changing of nuclear calcium level was monitored by laser confocal microscope.
