缩醛磷脂是一类甘油骨架的 sn-1 位置上通过烯醚键连接碳链的磷脂。缩醛磷脂在细胞生物学中发挥着多种重要的生物学功能,其水平紊乱与多种疾病的发生、发展密切相关。 由于同分异构体与同重素的存在给缩醛磷脂的质谱分析带来了一定的困难, 目前还未有一套完整明确的缩醛磷脂质谱分析方法。本课题将光化学反应——Paternò–Büchi反应与质谱技术相结合,采用微流反应器作为在线 PB 反应装置,在低能量碰撞诱导解离-串联质谱下获得可用于确定缩醛磷脂分子结构的特征诊断离子,有效区分缩醛磷脂与其他同分异构
体、同重素, 实现了对缩醛磷脂灵敏而快速的质谱分析。同时,在探索反应时间、溶剂组成、副反应等实验因素对 PB 反应产率影响的基础上,致力于提高缩醛磷脂PB反应的产率。

Plasmalogen is a subclass of glycerophospholipid characterized by the presence of a vinyl ether bond at the sn-1 position of the glycerol backbone. Plasmalogen is believed to play important biological functions in cell biology. Furthermore, recent research has shown that aberrant levels of plasmalogens are correlated with the onset and progression of many types of disease. The presence of
isomers and isobars brings difficulties to the analysis of plasmalogens via mass spectrometry (MS) and currently there is no single suitable method for unequivocal identification of plasmalogens. In this work, we aim to develop a novel method that
can achieve sensitive and fast identification of plasmalogen from other isomers and isobars through coupling the Paternò–Büchi reaction with CID-MS/MS. To achieve this goal, we studied the experimental parameters including reaction time , solvent
composition and side reactions to improve the PB reaction yield.
