秀丽隐杆线虫(Caenorhabditis elegans)在逆境下会产生休眠期幼虫,这一过程与其滞育机制密切相关。研究发现休眠相关基因(daf)在此过程中发挥重要的作用。松材线虫(Bursaphelenchus xylophilus)在逆境下也产生休眠期幼虫,但其机制尚不明确。本研究采用饲喂法用含有daf基因干扰载体(PHD-RH-daf)的灰霉饲喂松材线虫,观察并记录其发育速率、繁殖量和脂肪沉积量,以探讨daf基因在松材线虫休眠过程中的作用。此外,daf-8基因在休眠过程中可能起到潜在的重要作用。本研究克隆并分析了松材线虫daf-8基因的部分序列特征。主要结果如下: (1) 以PHD-RH空载为对照,干扰daf-16-1基因的松材线虫的发育显著降低(P<0.05);干扰daf-12和daf-16-1基因的松材线虫的繁殖量显著降低(P<0.05);干扰daf-3、daf-9、12、16-2基因的松材线虫脂肪沉积量明显增加。 (2) 松材线虫daf-8基因CDS序列全长为1626bp,编码541个氨基酸。通过对比加拿大(CAN)、浙江嵊泗(ZJSS)、印第安纳(IN1)、广东定海(DHBM)、浙江舟山(ZJZS)、日本(JAP)等六个地区来源的线虫daf-8基因序列,在其CDS区,共发现32个SNP位点,其中5个发生错义突变。同源性对比发现,ZJZS和JAP之间的序列同源性最高,为99.88%;CAN和IN1之间的同源性最低,为98.40%。

In unfavorable conditions, C. elegans undergoes development to dauer formation, which is closely related to the molecular basis of diapause and dauer formation genes (daf genes). Bursaphelenchus xylophilus undertakes dauer formation in unfavorable conditiongs as well, but its mechanism is not clear now. To make out how daf genes work in the diapause mechanism, we fed Botrytis cinerea that contains the interference vector PHD-RH-daf and analsized the development speed, the reproduction amount, as well as lipid accumulation. In addition, we cloned and analyzed the CDS sequence of daf-8 gene for further study on the potential founction in dauer formation in B. xylophilus. The main results are as follows: (1) Comparing to control of empty vertor, the developing rate of the nematode is significantly lower by RNA interference of daf-16-1 gene; the reproduction is significantly lower by RNA interference of daf-12 and daf-16-1 gene; and the lipid accumulation is significantly higher RNA interference of daf-3, daf-9, daf-12, daf-16-2 gene RNAi. (2) The CDS sequence of B. xylophilus daf-8 gene is 1626bp and can code 541 amino acids. There are 32 SNPS in the six geographic populations’ daf-8 gene CDS, which come from CAN, ZJSS, IN1, DHBM, JAP and ZJZS. Five of the 32 SNPS are missense mutations. The homology comparison reveals that the sequence homology is 99.88% between ZJZS and JAP, which is the highest among the six CDS. Also, there is a lowest homology of CDS sequences between CAN and IN1, which is 98.40%.
