胰腺星形细胞位于腺泡之间,一般处于静止状态,酒精及其代谢产物、氧化压力、细胞因子的刺激可导致其激活。本文分离鉴定了大鼠胰腺星形细胞。研究发现ATP刺激嘌呤能受体引发胰腺星形细胞胞浆钙离子浓度增加。胆囊收缩素(cholecystokinin, CCK)和乙酰胆碱(acetylcholine, ACh)对大鼠胰腺星形细胞胞浆钙离子浓度没有影响。新鲜分离的大鼠胰腺星形细胞存在自发钙振荡现象,这种自发钙振荡依赖于外钙,可被L型钙通道完全阻断。胰腺星形细胞显著表达CCK1受体。免疫组化、接近连接法等法证实CCK1受体与NADPH氧化酶偶联,CCK1受体的激活导致活性氧的产生。

Pancreatic stellate cells located around the acinus are usually in a quiescent state but alcohol and metabolites, oxidative stress, and cytokines stimulats its activation. We have isolated and identified rat pancreatic stellate cells investigated their regulation. It was found that ATP stimulation of purinergic receptors lead to increases in cytosolic calcium in pancreatic stellate.cells, but cholecystokinin (CCK) or acetylcholine (ACh) had no effect on cytosolic calcium in these cells. Freshly isolated rat pancreatic stellate cells exhibited spontaneous calcium oscillations which were eliminated by calcium-free extracellular solution and by the L-type calcium channels blocker nifedipine, but conotoxin GVIA, agatoxin, mibefradil had no effect. Immunocytochemstry demonatrated the presence of CCK1 receptors in pancreatic stellate cells, DuoLink proximity ligation asssy confirmed CCK1 receptor and NADPH oxidase association and interaction. The activation of CCK1 receptor leads to the generation of reactive oxygen species.
