苯并中等环系骨架作为重要的结构广泛存在于多种具有生物活性化合物中,发展构筑苯并中等环系骨架结构的新方法具有重要意义。氮杂环卡宾(NHC)作为一种重要的有机催化剂,为高效构筑多种手性结构提供了方法。然而,目前大量研究集中于构建五、六元环,利用NHC催化构建苯并中等环系的研究相对较少。本论文研究了NHC催化构建苯并中等环系骨架结构,分为两个部分,第一部分简单中介绍了中环化合物的结构和性质,并简要回顾了近年来关于中环化合物合成方法的研究进展;接着介绍了NHC的催化模式,并分析了NHC在催化不对称合成中环方向的应用。第二部分研究了以1-(2-氨基苯基)-2,2,2-三氟乙酮及肉桂醛为底物,使用NHC催化结合环扩张策略,设计合成了构筑苯并中等环系骨架结构的策略,并且初步探究了多种条件对该反应的影响。该合成方法有望拓展NHC有机催化剂的应用范围,丰富手性中环化合物的合成策略。

Benzo-fused medium ring skeletons exist widely in various biologically active compounds as important structures. As a result, it is of great significance to develop new methods for the construction of benzo-fused medium ring skeletons. As an important organocatalyst, N-Heterocyclic Carbene (NHC) provides a method for the efficient construction of various chiral structures. However, a large number of current researches about NHC focus on the construction of five- and six-membered rings, and relatively few studies on the construction of benzo-fused medium ring skeletons. This paper studies the NHC-catalyzed construction of benzo-fused medium ring skeletons, which is divided into two parts. In the first part, the structure and properties of the medium-sized rings are introduced, and the research progress on the synthesis methods of the medium-sized rings in recent years is reviewed. Then, the catalytic role of NHC is introduced, and the application of NHC in the catalytic asymmetric synthesis of medium-sized rings was analyzed. In the second part, we describe the design and synthesis of a benzo-fused medium ring framework structure, which is catalyzed by NHC and combined with ring-expansion strategy, with 1-(2-aminophenyl)-2,2,2-trifluoroethanone and Cinnamaldehyde used as the substrates. The effects of various conditions on the reaction are preliminarily explored. This synthesis method expands the application scope of NHC as organocatalysts and enriches the synthesis strategies of chiral medium-sized rings.
