马铃薯块茎蛾(Phthorimaea operculella (Zeller)),隶属于鳞翅目(Lepidoptera)、麦蛾科(Gelechiidae),是世界广泛分布的、对马铃薯(Solanum tuberosum)和烟草(Nicotiana tabacum)有严重危害的重要害虫。本文对马铃薯块茎蛾的生物学特征做了初步观察,对淡紫拟青霉(Paecilomyces lilacinus)、刀孢蜡蚧菌(Lecanicillium psalliotae)、与渐狭蜡蚧菌(Lecanicillium attenuatum)、蜡蚧轮枝菌(verticillium lecanii)、爪哇棒束孢(Isaria javanica)5种虫生真菌对马铃薯块茎蛾的卵、4龄幼虫及蛹的寄生作用进行了比较分析,并对筛选出的高毒菌株的生物学特性和致病效果进行了初步的测定,主要研究结果如下: 1.在实验室27℃,12L:12D,80%~90%的湿度条件下养殖并马铃薯块茎蛾对其生物学进行了观察,从卵发育到成虫大致需要25天,成虫羽化后4~7天产卵数量较多。 2. 筛选出了对该种害虫毒力更高的菌株爪哇棒束孢,测定出该菌对卵、4龄幼虫的半数致死浓度为2.5×10^6个/mL、0.85×10^6个/mL,在孢子浓度为1×10^7个/mL时,对卵、4龄幼虫、蛹的致死率分别为80.33%、72.21%、65.56%,对害虫寄生作用显著。 3. 不同的培养条件对爪哇棒束孢的生长和产孢有影响,菌落形态也有差异。 (1) 生长温度方面,爪哇棒束孢在33℃及以上,12℃及以下,菌落生长缓慢,27℃时生长最快; (2) 营养条件方面,在PDA、SMA、SDA、CMA、Czapek 五种培养基上,生长差异不显著; (3) 在PDA培养基上,光照和较高温度(30℃)下能够有效促进产孢,27℃及以下产孢很少甚至不产孢;黑暗条件下25℃下,五种培养基上PDA培养基上产孢很少甚至不产孢。

As an invasive species distributed widely around the world, Phthorimaea operculella had exerted a great deal of harm on the Solanum tuberosum, Nicotiana tabacu and other solanaceae crops and it was listed in the international and domestic important quarantine objects of transportation commercial. In this paper, five kinds of entomopathogenic fungi (Paecilomyces lilacinus、Lecanicillium psalliotae、verticillium lecanii、Isaria javanica、Lecanicillium attenuatum) were used to study the parasitic effects of the different stage of P. operculella: The egg stage,4th instar larva and pupa. The results provide a theoretical basis for the further development of biological control of P.operculella and the r&d of highly effective biological pesticides. The main contents are listed as following: 1 In the lab, under the condition of 27 degC,12 D: 12 L,and the humidity of 80% ~ 90% , using the potato tuber to breed the P.operculella and observing its biological characteristics. From egg to adult generally need 25 days and there are more eggs during 4 ~ 7 days after the adult eclosion. 2 Results showed that I. javanicus was more virulent to P.operculella of different stage, The corrected mortality of the egg stage, 4th instar larva and pupa were 80.33%,72.21%,65.56% respectively under 1 ×10 ^ 7 / mL concentration of the spore, The fungi to egg medium lethal concentration is 2.5 × 10 ^ 6 / mL,and 0.85×10^6/mL to 4th instar. 3 Different condition of culture affect the growth rate and the yield of the spore, and the colony morphology is different, too. (1)Under the different temperature conditions, the growth rate of the I. javanicus was notable discrepancy, and the optimum growth temperature was 27 degC, when the temperature was greater than 33degC or less than 12degC, the growth rate was slower or stop growing. (2)In the aspect of nutritional condition, the he differences of the growth rate on PDA, SMA, SDA, CMA, and Czapek were not significant. (3)On the PDA medium, light and high temperature (30 degC) to be able to effectively promote the yield of spores, When the temperature is 27 degC and below ,the yield of spores were little or no .Under dark condition, 25 degC, the yield of spores on five kinds of culture medium on spore production in PDA medium, the yield of spores were little or no, too.
