冬凌草是一种具有广泛的抗肿瘤活性的中草药,已经有文献报道它具有温热协同抑制人膀胱癌细胞生长的作用,但并未说明其有温热协同作用的成分及其作用机理。本课题的目的是:探讨冬凌草的主要抗癌活性成分--冬凌草甲素、以及冬凌草甲素与温热协同,对人膀胱癌细胞生长抑制的诱导作用,揭示冬凌草中抗肿瘤的主要活性成分,为进一步研究冬凌草的抗癌机制提供依据。方法:用不同浓度的冬凌草甲素与人膀胱癌细胞作用,在不同的温热条件下协同处理,用MTT法检测药物对细胞的生长抑制作用。结果:冬凌草甲素与人膀胱癌细胞生长抑制有显著的量效关系,显示其具有体外抗肿瘤作用;同时观测到冬凌草甲素在43℃加热0.5h具有明显的热协同作用,而在41℃时,热协同作用不明显。另外比较发现,冬凌草单体(冬凌草甲素)的抗肿瘤能力不及冬凌草全草(水煎剂),说明冬凌草中还有其他的抗肿瘤活性成分,这些成分也极有可能具有热协同作用。该实验也佐证了:中药具有多靶点、多组分药理协同作用的理论。 |

It has been found that Rubdosia (a traditional Chinese medicine with anticancer function) has inhibitory effect and co-effect with hyperthermia on human bladder carcinoma, but the mechanism is not for sure. This paper aims to investigate the effect of Oridonin, which is the main anticancer component of Rubdosia, as well as the co-effect with hyperthermia on human bladder carcinoma using BIU-87 cell line in vitro. MTT method was chose to evaluate the inhibitory effect and co-effect combining with 0.5h cultured in 41℃ and 43℃ on BIU-87 cells growth at different concentrations of oridonin. Results: Oridonin has the inhibitory effect of human bladder carcinoma growth in vitro, farther more there is s a dose-response relationship between different concentrations of oridonin and human cancer cells inhibition; And a co-effect of oridonin with 0.5h hyperthermia at 43℃ was observed but not at 41℃; Comparing oridonin to rubdosia, the inhibitory effect of oridonin to human bladder carcinoma is inferior to rubdosia, showed that rubdosia has other anticancer components which would have inhibitory effect on human bladder carcinoma and co-effect with hyperthermia. |
